The Infinite Scroll cannot be used in homepage posts due to the design of the theme. If you select your homepage as a static page and then select your homepage and posts page the infinite scroll should work on the posts page or the blog page.

I checked the plugin with the Blankslate theme and the infinite scroll seems to be working with the default plugin selectors. By the looks of the image you seems to be using a plugin for the posts.
